Cardiac hypertrophy is a pathophysiological response to various pathological stresses and ultimately leads to heart failure. Oxidative stress is one of the key processes involved in hypertrophy development. Fisetin supplement(also known as fisetin, fisetin) is a small molecule flavonoid that has been shown to have antioxidant, anti-proliferative and anti-inflammatory properties. However, the effect of fisetin on cardiac hypertrophy is unknown.
In the current study, Bin Dong, Chen Liu and others from the Department of Cardiology, the First Affiliated Hospital of Sun Yat-sen University in China and the Key Laboratory of Assisted Circulation of the Ministry of Health found that fisetin inhibited cardiac hypertrophy caused by pressure overload, improved cardiac function in vivo and in vitro Inhibits phenylephrine (PE)-induced cardiomyocyte hypertrophy. Fisetin treatment significantly reduced reactive oxygen species (ROS) levels in hypertrophic hearts and cardiomyocytes.
In addition, fisetin significantly up-regulated the expression of antioxidant genes, including catalase (CAT), superoxide dismutase 1 (SOD1) and heme oxygenase 1 (HO-1). In addition, co-treatment of N-acetylcysteine (NAC, ROS scavenger) and fisetin had no synergistic inhibitory effect on PE-induced cardiomyocyte hypertrophy, indicating that the anti-hypertrophic effect of fisetin is mainly related to blocking oxidative stress. .
Finally, after pressure overload and PE treatment, pro-hypertrophic signaling pathways, mitogen-activated protein kinase (MAPK) and mammalian target of rapamycin (mTOR) kinase were found to be inhibited by fisetin.
